1) This medium can grow a wide variety of aerobic and anaerobic organisms, from fastidious to non-fastidious. 2) The band at the surface of the broth is resazurin, an oxygen indicator that turns pink when oxidized and remains colorless when reduced. It is more desirable to have a thin band because that would create a wider gradient, as less of the medium is fully oxidized. 3) strict aerobe- only at the top anaerobe- only at the bottom microaerophile- a band just below the surface facultative anaerobe- better growth at the top, but some growth throughout 4) The medium should be fresh so that there is less time for oxygen to diffuse throughout the broth. Old media would negatively affect the growth of anaerobic organisms (and possibly microaerophilic) because of the increased presence of oxygen. Growth of aerobes and facultative anaerobes would be affected positively.
